摘要与影响
CRISPR-Cas systems, originally discovered as a part of the adaptive immune system in bacteria and archaea, have revolutionized genetic engineering across various organisms, including insects and other arthropods. These systems enable precise genome editing by utilizing the Cas protein, guided by RNA, to target and modify specific DNA sequences. CRISPR-Cas technologies have been employed in insects and other arthropods to study gene functions, develop pest control strategies, and improve disease vector management. For instance, in mosquitoes, CRISPR-Cas has been used to disrupt genes essential for malaria transmission, offering a promising avenue for reducing the spread of this disease. In agricultural pests, genome editing facilitates the development of insect-resistant crops and environmentally friendly pest control methods. Model organisms like Drosophila melanogaster have been pivotal in advancing CRISPR research, enabling precise gene editing, functional genomics studies, and genetic screens. In agricultural pests, CRISPR-Cas offers targeted gene modifications for population control and resistance management, with innovations like gene drives and the Sterile Insect Technique. Beneficial insects such as bees and butterflies benefit from CRISPR-enhanced disease resistance, improved gut microbiomes, and modifications for better pollination efficiency. Additionally, CRISPR-Cas systems aid in unraveling the complex genetic mechanisms underlying behaviors, development, and physiology in diverse arthropod species. Despite the transformative potential of CRISPR-Cas, challenges such as off-target effects, delivery methods, and ethical considerations remain. This chapter highlights the significant advancements and future directions of CRISPR-Cas applications in insects and other arthropods, emphasizing their impact on both basic research and practical applications in biotechnology and agriculture.
